Well, I just came back from a trip down to Ushuaia and El Calafate, which I did BY BUS. OK, so first recommendation - you should really try to go by plane.

By Bus
So, we went down to El Retiro. This is where all the buses leave from and where all the companies sell you the tickets. We went with Andesmar and the service was good. Meals are included and they play a bunch of movies so you don't get that bored in the 48 HOUR ride!

TIP
Negotiate. Prices can be negotiated. Also, if you are studying in Argentina, make sure to bring your student ID card, cause it will get you a very STEEP discount. All in all, we ended up paying about $280 for the round trip bus ticket.
It's a 48 HOUR trip. So, I cannot emphasize hard enough that you should consider going by plane.
By Plane
Plane can be expensive if you try to buy it directly. The prices advertised are for Argentineans, and foreigners are charged a lot more. However, there is a way to get a much cheaper flight, which will run you about $100 to $200 more than the bus option - and that is by buying a package.
TIP
The cheapest I found was through Despegar.com. The package that makes the most sense is the one getting to El Calafate, which is where the glaciers are at. From there, you can coordinate a bus ticket to Ushuaia which can run you about $70 round trip.

To get to Ushuaia, there is no direct route from El Calafate. You will have to stop at Rio Gallegos and board another bus to Ushuaia.
